Further information: | The plasmid was created by introducing an Y395C mutation into the S. cerevisiae GAP1 coding sequence of the pGAP1-GFP vector. The plasmid contains the S. cerevisiae GAP1 promoter, the mutated GAP1 coding sequence in frame with the C-terminal mutated GFP coding sequence and the GAP1 terminator. The plasmid contains the S65T-GFP mutant, whose fluorescence is known to be pH sensitive (Kneen et al., 1998). |
