Cloned DNA: Vibrio fischeri luxR gene
Vibrio fischeri luxICDABEG operon consisting of the luxI, luxC, luxD, luxA, luxB, luxE and luxG genes
Promoter: Vibrio fischeri luxR promoter
Vibrio fischeri luxICDABEG operon promoter
Ribosome
binding site:
Ribosome binding sites (RBS) of the Vibrio fischeri luxR, luxI, luxC, luxD, luxA, luxB, luxE and luxG genes
Terminator: Vibrio fischeri luxICDABEG operon terminator
Selection marker: Ampicillin (amp)
Replicon: Escherichia coli plasmid pMB1 origin
Host range: Escherichia coli
Parental clone: pBR322
Further information: This plasmid was constructed by cloning a 9kb SalI fragment of V. fischeri MJ-1, containing the luxR gene and the luxICDABEG operon consisting of the luxI, luxC, luxD, luxA, luxB, luxE and luxG genes, into the SalI opened pBR322.
The luxR gene and the luxICDABEG operon are transcribed divergently, respectively from the luxR and the luxICDABEG operon promoter that are located in between the luxR and the luxI genes.
The lux promoters and RBS sites are not shown on the circular map of the plasmid.
pJE202 contains all functions required for spontaneous light emission; addition of external substrate is not necessary.
The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727306.1.
The nucleotide sequence of the inserted SalI fragment corresponds with the EMBL Nucleotide Sequence Database accession number AF170104.1.
