Cloned DNA: Influenza A virus matrix protein 2 cDNA (M2)
Glycine max cytosolic ascorbate peroxidase 1 cDNA (APX1, GeneID 553156); improved monomeric enhanced mutant (APEX2)
Promoter: Human cytomegalovirus immediate early promoter (CMV-IE) and enhancer
Simian virus 40 early promoter (SV40 early)
Escherichia coli β-lactamase promoter (amp)
Ribosome
binding site:
Ribosome binding site (RBS) of the Escherichia coli ampicillin resistance gene (amp)
Terminator: Herpes simplex virus (HSV) thymidine kinase polyadenylation signal (TK polyA)
Simian virus 40 polyadenylation signal (SV40 polyA)
Selection marker: Neomycin (neo; G418; kanamycin (kan))
Replicon: Escherichia coli plasmid pMB1 origin
Phage f1 origin
Simian virus 40 bidirectional origin (SV40)
Host range: Escherichia coli
Mammalian cells; SV40 permissive cells
Parental clone: pPTB
Further information: The plasmid was constructed by cloning the Influenza M2 and soybean APEX2 coding sequences into the pPTB vector.
APEX2, the second-generation monomeric enhanced APX mutant, contains an additional mutation (A134P) compared to APEX (K41D, W41F, E112K) and is used as a reporter to generate contrast for electron microscopy and also for proteomic tagging in living cells.
Other name of the plasmid is pM2Apex2.
