Cloned DNA: Mouse tumor necrosis factor (Tnf, TNF); fragment
Human tumor necrosis factor (TNF); fragment
Promoter: Phage λ major leftward promoter (λ PL)
Ribosome
binding site:
Ribosome binding site (RBS) of the phage Mu ner protein gene
Terminator: -
Selection marker: Ampicillin (amp)
Replicon: Escherichia coli plasmid pMB1 origin
Host range: Escherichia coli; use strains with a cI function, cIts for PL controlled expression
Parental clone: pPLcmuHTNF1; pATTrpMTNF
Further information: This plasmid was constructed as follows : the NcoI-BanI fragment of pATTrpMTNF (containing the first part of mature mouse TNF) was inserted into the NcoI-BanI sites of pPLcmuHTNF1.
pPLcmuMHTNF4 is designed for a PL-controlled expression of intracellular mature human-mouse hybrid TNF.
Restriction analysis revealed that the plasmid is approx. 1600 bp shorter as described. Supposedly the plasmid is derived from a pPLc245HTNF-plasmid and not from pPLcmuHTNF1. The nucleotide sequence file has not been adapted.
Other names of the plasmid are MH4 and pPLc245-MH4.
