| Further information: | The plasmid was constructed by inserting the BsgI (blunted) - SapI (filled in) fragment from pSCGAL1mBAX-L, containing the S. cerevisiae galactokinase promoter (GAL1), the α variant of the mouse Bcl2-associated X cDNA (mBAX) and the S. cerevisiae 2 micron plasmid (2μ) FLP terminator, into the Ecl136II opened pRS425 vector. pRS425GAL1mBAX is a high-copy Yeast Episomal Plasmid (YEp) for expression of the mouse Bax-α protein. When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide. The nucleotide sequence of the mBAX cDNA corresponds with the EMBL Nucleotide Sequence Database accession number L22472.1. Other names of the plasmid are pRS425GALmBAX and pRS425Bax. |
