| Further information: | This plasmid was derived from pSCTPIMF1 as follows: the SalI-EcoRI fragment of pSP64GAL1m (containing the galactokinase (GAL1) promoter) was inserted into the XhoI and EcoRI sites of pSCTPIMF1. Expression sites: HindIII and StuI. The BamHI site at the 3' end of GAL1 is lost; the sequence in this region is not exactly known. ATTENTION: the vector part of the plasmid is probably not correct: restriction sites were not experimentally verified. Because of the presence of the lac operator, Lac+ E. coli host cells turn blue on X-Gal. When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide. Other name of the plasmid is pSCGal1MF2. |
