Further information: | The unique StuI site at the end of the prepro sequence was introduced by mutation . The segment from residue 149 until 401 corresponds in anti- clockwise orientation to the 3' end of the lacZ alpha fragment. ATTENTION : the vector part of the plasmid is probably not correct: the restriction sites were not experimentally verified. Usable fragment: EcoRI-StuI fragment or BglII-StuI fragment. Only the first spacer sequence (to HindIII) is on the plasmid. Because of the presence of the lac operator, lac+ E. coli host cells turn blue on X-Gal. When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide. . The nucleotide sequence from nucleotide position 1620 to 1320 corresponds with the upstream sequence of the yeast α-mating factor 1 (MF) promoter. Other name of the plasmid is pMATA2151H2. |
