Further information: | pUC8 is similar to pUC18, but contains fewer cloning sites in the lacZα region. Designed as an insert detection vector using α-complementation recently used for the expression of fused proteins: ATG in phase with GTC from E-AccI, E-HindII, E-SalI. ATG in phase with CGG from E-AvaI and E-SmaI. ATG in phase with GGA from E-BamHI. ATG in phase with CTG from E-BspMI. ATG in phase with AAT from E-EcoRI. ATG in phase with AGC from E-HindIII. ATG in phase with CTG from E-PstI. pUC8 has no PstI site in the ampicillin resistance gene. When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide. The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database entry (Release 37; Accession number L08959), starting at another nucleotide position and complemented. |
