Further information: | This plasmid was derived from pSP65HIL4f2 (the same plasmid as pSP65HIL4f but with hIL4f in the clockwise orientation) as follows: The EcoRI site near the SP6 promoter was converted to an Asp700 (XmnI) site by partial digestion and filling in with Klenow DNA polymerase, resulting in pSP65HIL4f3 having a single EcoRI site present in the coding sequence of mature hIL4. In this EcoRI site the second EcoRI fragment of the EcoRI linked hIL4 cDNA (from the same λgt10 phage) was inserted (the second EcoRI linker was placed approx. 90 nucleotides behind the termination codon of the hIL4 gene). When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide. The nucleotide sequence of the hIL4 cDNA was assumed to be identical to the sequence reported by Yokota et al. (1986). |
