Further information: | The plasmid was constructed by changing 16 codons to glycine or alanine codons in the IVNA coding sequence of pSV23IVNAm2, using the site-specific mutagenesis protocol described in Morinaga et al. (1984). This plasmid was constructed to investigate the protective potential of neuraminidase mutants towards different influenza strains. The IVNA coding sequence contains the following mutations: C42S, D43M, S44A, C78R, a silent mutation in codon 79, K328A, N329G, R331A, S334G, T336A, N339G, K344A, a silent mutation in codon 345, N346A, H347A, S367A, E368A, D369G, S400A, A401G, R403G and E431G. The additional mutations compared to pSV23IVNAm2 give rise to additional SacII and ApaI sites, and 2 additional BamHI restriction sites. The nucleotide sequence of wild type IVNA corresponds with the EMBL Nucleotide Sequence Database accession number CY015510.1. Other name of the plasmid is pSV23IVdNAN5. |
