Further information: This plasmid was constructed as follows: the small HindIII-FnuDII fragment of pPLaIVNA (containing the first 45 codons of Influenza A/Victoria/3/75 neuraminidase gene) was isolated, provided with a Gly-Ser-Ser, BglII linker, and inserted between the HindIII and BglII sites of pSV23SmTNFm3. This results in the fusion of the neuraminidase anchor to mature mTNF.
The plasmid contains the SV40 small t-antigen splicing signal.
Restriction enzyme digestions revealed that approx. 60 nucleotides are missing from the 3' untranslated region of the mTNF gene, including the 3'-terminal (supposedly restored) EcoRI site.
The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727483.1.
