Further information: The plasmid was constructed as follows: pSV51B1sFvHG3f was cut with BspMII/EcoRV (partial BspMII digestion at nucleotide position 4651), and also cut, in a second reaction, with EcoRV/XbaI. 2) The resulting 1062 bp BspMII/EcoRV fragment was then ligated to the 7889 bp EcoRV/XbaI fragment. 3) The 749 bp NheI/BspMII fragment of pQEB1sFv could then be inserted. As a result, the hinge region and CH2 and CH3 domains of the human immunoglobulin γ3 heavy chain (HG3f) were eliminated.
The C-terminal polyhistidine tag permits detection and purification of the recombinant B1sFv protein.
After transient expression in COS-1 cells, the B1sFv protein was secreted into the culture medium.
