Further information: | The plasmid was constructed by cloning the SV40 large T-antigen from pMaSVT into the EcoRI/XbaI opened pT2/BH vector. This plasmid enables easy immortalization of almost any cell by co-transfection of pT2-SVT and Sleeping Beauty transposase (SB100X). This is far more efficient than transfection of linearized pMaSVT. From a biosafety perspective, it is still as safe as any normal plasmid, in contrast to using the SV40 large T-antigen in for example a lentivirus, which carries a significant biosafety hazard. |
