Further information: The plasmid was constructed by ligating the EcoRI-digested pIL253 vector to the EcoRI-digested pSA34 vector.
pTRKH1 is a shuttle cloning vector for E. coli, Lactococcus, Enterococcus, Streptococcus and Lactobacillus species.
The vector has a medium copy number (30-40) in E. coli, and a high copy number (45-85) in streptococcal and lactococcal hosts.
Tetracycline resistance is only expressed in E. coli, erythromycin resistance is expressed both in E. coli and in Gram-positive bacteria.
The tetracycline resistance gene can be insertionally inactivated, facilitating screening for clones.
