| Further information: | This plasmid was constructed by inserting an approximately 1500 bp EcoRI fragment of λ gt10 mIL5-1 (containing mouse Il5 cDNA) into the EcoRI site of pUC13. The orientation of the insert (sense relative to the lac promoter) was verified by a NdeI and a SspI restriction site analysis. When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide. The nucleotide sequence of the mouse Il5 cDNA corresponds with the Genbank accession number NM_010558.1, except for the adjustments due to the sequence analysis results. The mouse Il5 coding sequence contains 2 silent mutations, one at codon position 37 (S37S; TCC->TCT) and one at codon position 127 (T127T; ACA->ACG), and 1 non-silent mutation at codon position 79 (R79H; CGT->CAT), compared to the reference sequence NM_010558.1. Other name of the plasmid is pUC13mIL5c. |
