Further information: The plasmid contains the Candida albicans codon adapted coding sequence of the mouse Bcl2-associated X cDNA (CaBAX), inserted into the polylinker of pUC21.
The plasmid is non-transferable and has a high copy number.
The strength of the lac promoter is reduced by increasing the distance between the -10 and -35 elements: the HpaII site was filled in, creating a SacII site.
The yield of plasmid ssDNA is improved by a deletion within the M13 minus ori (from nucleotide 5644 to 5721 of the M13 genome).
When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide.
