Further information: | The plasmid contains the Candida albicans codon adapted coding sequence of the mouse Bcl2-associated X cDNA (CaBAX), inserted into the polylinker of pUC21. The plasmid is non-transferable and has a high copy number. The strength of the lac promoter is reduced by increasing the distance between the -10 and -35 elements: the HpaII site was filled in, creating a SacII site. The yield of plasmid ssDNA is improved by a deletion within the M13 minus ori (from nucleotide 5644 to 5721 of the M13 genome). When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide. The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727208.1. The nucleotide sequence of pUC21 corresponds with the Genbank accession number AF223641.1. |
