| Further information: | The coordinates of the plasmid are not those as used by Ruther but have been rearranged to fit the coordinates of the pUC vectors. pUR250 was designed as an insert detection vector using α-complementation of the lacZ gene. Although the plasmid can be used as an expression vector using lac, the pUC series are more versatile for this purpose. When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide. Caution: the plasmid was mutagenised in vivo and not resequenced. It may therefore contain unknown mutations. |
