| Further information: | To make this plasmid pUR250IVHA8s was BamHI opened (at the 3' end of the heamagglutinin gene, partial digestion), filled in with Klenow DNA polymerase, and a linker coding for stops in all three reading frames (CT TAG T TAA C TAA G) was inserted. Only two amino acids are added by the STOP linker insertion; Leu (amino acid 187) identical to the wt sequence and Ser. This plasmid is a basic construction for all HA8si containing vectors. When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide. Other names of the plasmid are pUR250H8A-STOP and pUR250HA8STOP. |
