Further information: pURgHIL22 contains the third and fourth exon of the human interleukin 2 gene (hIL2), part of the second intron (approx. 1000 bp of a total of 2300 bp) and the third intron.
The sequence of the genomic hIL2 insert was obtained from Genbank 'Locus: HUMIL2'; it differs in several nucleotides from the incompletely determined hIL2 sequence as described by Degrave et al. (1983); more differences are to be expected, since the XbaI site at position 3220 (i.e. almost coinciding with the unique BglII site in intron 3) in the genomic hIL2 sequence obtained from Genbank and described by Fujita et al. (1983) could not be detected experimentally in the clone pURgHIL22 of Degrave et al. (1983). Note that, because of the computer plot programme, this hypothetical XbaI site is indicated on the circular map.
When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide.
The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727567.1.
