Further information: The plasmid was constructed by removing the Ig2 from pcDNA3-F-HLH-Ig2-VSV-MALT1C-HA as a BamHI/XhoI fragment, and replacing it with a BglII-KpnI-BamHI-Ig1-Ig2-XhoI fragment. The resulting HLH-p76 MALT1 is in frame with the VSV-MALT1C-HA.
The plasmid is intended for functional characterization of mutations and (auto)processing events in human MALT1, via strategically placed FLAG, VSV and HA epitope tags.
The plasmid contains the ETV6 helix-loop-helix domain (HLH) which is a dimerization domain, shown to strongly activate human MALT1.
The immunoglobulin-like domains Ig1 and Ig2 of hMALT1 contribute to the oligomerization of the caspase-like domain and help with the correct dimerization.
The HLH domain can be deleted using KpnI.
