Further information: | The plasmid was constructed by digesting the plasmid pTRKH1 with AvaI and religating, with loss of two small AvaI fragments. pTRKH3 is a shuttle cloning vector for E. coli, Lactococcus, Enterococcus, Streptococcus and Lactobacillus species. The vector has a medium copy number (30-40) in E. coli, and a high copy number (45-85) in streptococcal and lactococcal hosts. Tetracycline resistance is only expressed in E. coli, erythromycin resistance is expressed both in E. coli and in Gram-positive bacteria. The tetracycline resistance gene can be insertionally inactivated, facilitating screening for clones.
