Further information: | The plasmid was constructed as follows: the Sau3AI fragment of genomic DNA of C. albicans B2630, containing the MET15 gene, was inserted into the BamHI site of YCp50. To shorten the genomic DNA fragment, the plasmid was digested with EcoRI and the largest fragment was religated, eliminating also 377 bp of YCp50. The nucleotide sequence of the CaMET15 genomic DNA corresponds with the EMBL Nucleotide Sequence Database accession number AF188275.1. Referring to Viaene et al. (2000), sequence analysis of the genomic CaMET15 insert at the Department of Biomedical Molecular Biology (Ghent University, Belgium) revealed the presence of 1320 bp coding sequence, 542 bp upstream and 314 bp downstream sequence. According to the EMBL record AF188275 and the Candida genome (http://www.candidagenome.org/) however, 315 bp downstream sequence (incl. termination codon) is present. |
