Further information: | The plasmid was constructed by replacing the 5' CaADH1-TDH3 and 3' CaADH1 regions in pNIM1T-tTA with the 5' and 3' regions of CaRPS1. The plasmid is intended for chromosomal integration of the GFP-nourseothricin resistance cassette into the C. albicans genome via recombination with the RPS1 promoter and terminator sequences. Integration allows for tetracycline/doxycycline regulated expression of GFP via a Tet-Off transactivator, under the control of the C. albicans RPS1 promoter. The coding sequence of GFP was codon optimised for expression in C. albicans. The nourseothricin resistance gene CaSAT1 consist of the N-terminal part of the C. albicans ACT1 gene (intron included) fused to the E. coli SAT1 gene, codon optimised for C. albicans. The nucleotide sequence of the plasmid corresponds with the EMBL Nucleotide Sequence Database accession number KX766193.1. |
