Further information: The plasmid was constructed by cloning the GFP coding sequence, under control of the tet-regulated promoter, into the pNIMXT-rtTA vector.
The plasmid is intended for chromosomal integration of the GFP-nourseothricin resistance cassette into the C. albicans genome via recombination with the 5' and 3' ADH1 sequences.
Integration allows for tetracycline/doxycycline regulated expression of GFP via a Tet-On transactivator, under the control of the C. albicans TDH3 promoter.
The coding sequence of GFP was codon optimised for expression in C. albicans.
The nucleotide sequence of the plasmid corresponds with the EMBL Nucleotide Sequence Database accession number KX766190.1.
