| Further information: | The plasmid was constructed by inserting the 1950 bp XhoI fragment from pCAGGS-Flag-hABIN1, containing the human A20-binding inhibitor of NF-κB activation 1 (hABIN-1) mutated sequence, into the XhoI opened pSP72 vector, in the sense orientation relative to the T7 promoter. As compared to the wt hABIN-1 cDNA, the proline (P) codon at position 299 is replaced by an alanine (A) codon resulting in the loss of an AvaI site and the isoleucine (I) codon at position 9 contains a silent mutation (atc -> att) resulting in the loss of a DpnI and a Sau3AI site. The nucleotide sequence of the pSP72 vector was obtained from Genbank (Accession number X65332.2), corresponding to the Promega website (version 12/01/2000). The nucleotide sequence of the hABIN-1 cDNA was obtained from Genbank (Accession number NM_006058.2). Other name of the plasmid is pSP72-hABIN1S9. |
