Further information: | The plasmid was constructed by changing 3 codons to glycine or alanine codons in the IVNA coding sequence of pSV23IVNAm2, using the site-specific mutagenesis protocol described in Morinaga et al. (1984). This plasmid was constructed to investigate the protective potential of neuraminidase mutants towards different influenza strains. The IVNA coding sequence includes the following mutations: C42S, D43M, S44A, C78R, a silent mutation in codon 79, S400A, A401G and R403G. The additional mutations compared to pSV23IVNAm2 give rise to an extra BamHI restriction site. The nucleotide sequence of wild type IVNA corresponds with the EMBL Nucleotide Sequence Database accession number CY015510.1. Other name of the plasmid is pSV23IVdNAN4. |
