Further information: | The plasmid was constructed by changing 3 codons to glycine or alanine codons in the IVNA coding sequence of pSV23IVNAm8, using the site-specific mutagenesis protocol described in Morinaga et al. (1984). This plasmid was constructed to investigate the protective potential of neuraminidase mutants towards different influenza strains. The IVNA coding sequence includes the following mutations: C42S, D43M, S44A, C78R, a silent mutation in codon 79, D147G, T153G, a silent mutation in codon 154, H155G, a silent mutation in codon 196, Y197A, K199G, K328A, N329G, R331A, S334G, T336A, N339G, K344A, a silent mutation in codon 345, N346A, H347A, S367A, E368A, D369G, S400A, A401G, R403G, E431G, D463G and L466G. The nucleotide sequence of wild type IVNA corresponds with the EMBL Nucleotide Sequence Database accession number CY015510.1. Other name of the plasmid is pSV23IVdNAN8. |
