| Further information: | The plasmid was constructed as follows: 1) The E6Hf1 gene was first cloned as a HindIII-SalI fragment from pSV23SE6Hf1 into pPLcT73, resulting in the intermediate construction pPLcT73E6Hf1; 2) The E6Hf1 gene was then cloned as an XbaI fragment from pPLcT73E6Hf1 into the XbaI opened pSV51 vector (base position 3087 and 3636). The plasmid is similar to pSV51E6H, exept for the cloned gene. While cloning the E6Hf1 gene as an XbaI fragment into pSV51, the short 3' flanking sequence of the E6Hf1 gene was cut off, because the HindIII-SalI fragment contains an internal XbaI site close to the SalI site. The plasmid is used for the expression of the truncated heavy chain of the Fab fragment E6F1 (E6Hf1) of the mouse anti-hPLAP monoclonal antibody E6(IgG2b,κ). The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727622.1. |
