Further information: The plasmid was constructed by introducing an R149Q mutation into the human MALT1 fragment of pcDNA3.1-Flag-hMALT1-N via site-directed mutagenesis.
This plasmid contains an N-terminal fragment of human MALT1, encoding the DD-Ig1-Ig2 domains. This fragment acts slightly dominant-negative downstream of CARMA1 overexpression in NF-kB luciferase assays, indicating that it can compete with endogenous MALT1 for critical adaptor proteins.
The R149Q mutation corresponds with the human SNP rs147414021, which involves an autoprocessing site in MALT1.
The nucleotide sequence of the wild type human MALT1 cDNA corresponds with the EMBL Nucleotide Sequence Database accession number AF130356.2.
