Further information: | The plasmid was constructed by cloning a fragment of the phage T7 genome, containing the T7 lysozyme coding sequence, into the BamHI-opened pACYC184 vector. This plasmid is intended for co-transformation with a bacterial expression vector in E. coli BL21(DE3) (LMBP 1455). The phage T7 DNA is expressed via the tet promoter, consequently pLysE (Expressed) supplies a high level of phage T7 lysozyme. This level of lysozyme reduces the fully induced activity of T7 RNA polymerase such that induced cells can continue to grow and produce innocuous target proteins indefinitely. |
