Further information: The plasmid was constructed as follows: 1) a cDNA library from Arabidopsis thaliana was constructed in pUC18; 2) from all SOD containing clones, the one with the largest but still 5'-incomplete A. thaliana iron superoxide dismutase cDNA (FeSOD) insert was selected.
The FeSOD insert encodes 23 amino acids at the 5' end, which could be part of a chloroplast transit sequence. Amino acid 24 (asparagine) is presumably the first of the mature protein. Genbank Accession number NM_118642 marks a shorter transit sequence (only 10 codons).
When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide.
The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727070.1.
The nucleotide sequence of the FeSOD cDNA corresponds with the Genbank accession number M55910.1.
