| Further information: | The plasmid was constructed as follows: pSP65HIL4 was XmnI opened, BglII linkers (5'-CAGATCTG-3') were added and the BglII-BamHI fragment (containing the hIL4 cDNA) was inserted into the unique BglII site of pSP64GMT. The expression level obtained after injection of transcripts in oocytes was ten times higher than for transcripts of pSP65HIL4. When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide. |
