Further information: The plasmid was constructed as follows: the hTNFR75 coding sequence was inserted as a HindIII-Asp718 (KpnI) fragment from pSPT1hTNFR75 into the unique HindIII and Asp718 (KpnI) sites of pSV25S.
The eukaryotic Kozak recognition sequence (GCCGCCACC ATG G) for initiation of translation of hTNFR75 is present.
The plasmid contains the SV40 small t-antigen splicing signal.
Name of the plasmid in Vandenabeele et al. (1995) is pSV25S-hTNF-R75.
The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727530.1
The nucleotide sequence of the human TNFR75 cDNA corresponds with the EMBL Nucleotide Sequence Database accession number M32315.1. However, due to the PCR amplification for the construction of the parental clone pSPT1hTNFR75, there is one base pair exchange in one codon of the hTNFR75 coding sequence that does not affect the amino acid sequence (silent mutation; W. Lesslauer, F. Hoffmann - La Roche LTD, Basel, Switserland: personal communication). This mutation was not experimentally verified by BCCM/GeneCorner.
