Further information: This is a recombinant expression vector for canine interferon γ under control of the SV40 early promoter.
The genomic canine interferon γ gene was inserted into pSV23 as two fragments: 1) a 750 bp RsaI-EcoRI fragment, with SalI linker ('CGTCGACG') ligated to the blunt RsaI, was isolated from subclone pPL-gcIFN-γa, and was then cloned into the SalI-EcoRI opened pSV23 vector. The sequence of the fragment encompasses the mRNA transcription initiation site up to the EcoRI site located in the first intron. This provided the intermediate construction pSVgcIFN-γ ExI, lacking a SV40 polyadenylation signal. 2) A 5 kb EcoRI fragment, isolated from subclone pgHIFN-γ Eco5, containing exons II, III and IV as well as the polyadenylation signal, was cloned into pSVgcIFN-γ, opened with EcoRI.
Biological activity was shown in CHO-transfected cells.
Since the nucleotide sequence is not completely known, there is uncertainty as to the cutting frequency of the restriction enzymes indicated.
Name mentioned in Devos et al. (1992) is pSVgcIFN-γ.
Other name of the plasmid is pSVd2-3gCIFNγ.
