Further information: The plasmid was constructed by inserting the Candida albicans O-acetylhomoserine O-acetylserine sulfhydrylase (CaMET15) gene as a Sau3AI/EcoRI fragment of YCpCaMET15 (sites filled-in with T4 DNA polymerase) into the SmaI opened pRM2 vector.
The presence of both C. albicans autonomously replicating sequences ARS2 and ARS3 allows direct recovery in Escherichia coli of non-multimerized plasmids from C. albicans transformants without any intermediate step in Saccharomyces cerevisiae.
When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide.
The nucleotide sequence of the CaMET15 genomic DNA corresponds with the EMBL Nucleotide Sequence Database accession number AF188275.1.
The nucleotide sequence of the CaURA3 DNA corresponds with the EMBL Nucleotide Sequence Database accession number X14198.1. HOWEVER, THERE MIGHT BE CORRESPONDENCE WITH AF109400.1: see LMBP 4100.
