Further information: The plasmid was constructed by cloning the HindIII-SalI fragment from pATcE6Hy3 into the unique E-HindIII and E-SalI sites of pSV23.
This plasmid contains a complete cDNA insert encoding the complete mouse::human chimeric heavy chain of the anti-hPLAP antibody E6(Hy3,κ) clockwise orientated relative to the SV40 early promoter.
The chimeric CH1 domain contains 60 amino acids of mouse (E6H) and 37 amino acids of human (hIG3) origin.
The plasmid doesn't contain the small t-antigen splicing signal.
Since the prokaryotic part is derived from pAT153, there are no 'poison' sequences present.
Designed for transient expression in COS cells or constitutive expression in CHO cells (cotransfection with DHFR plasmids).
The nucleotide sequence of the SacI-NsiI fragment of pOMMB was obtained from Genbank 'Release 55; Locus : HUMIGHAF' and differs in 6 nucleotides from the sequence of the coding regions of the E6Hy3 SacI-NsiI fragment of pATE6Hy3. The unknown nucleotides in the Genbank-obtained sequence have been sequenced (Dr R. Schoonjans, Department of Biomedical Molecular Biology, Ghent University, Belgium).
