Further information: E6Hy3f2 is a HindIII-HindIII fragment of pATE6Hy3; the second HindIII site is made by filling in (Klenow polymerase) the unique MstII site at nucleotide position 1954 of the E6Hy3 gene in pATE6Hy3, and adding at least 2 multi-stop-linkers: 'CTTAGTTAACTAAG'; the succession of the two linkers resulted in the formation of this second HindIII site.
The multi- stop-linker contains a HpaI site.
This plasmid contains a half-genomic insert encoding the truncated mouse::human chimeric heavy chain of the anti-hPLAP antibody E6(Hy3,κ) clockwise orientated relative to the SV40 early promoter. The mouse part of the cloned gene consists of cDNA; the human part consists of genomic DNA.
The plasmid doesn't contain the SV40 small t-antigen splicing signal.
The truncated CH2 domain consists of 31 amino acids, terminating at the multi-stop-linker.
Designed for transient expression in COS cells or constitutive expression in CHO cells (cotransfection with DHFR plasmids).
