Further information: The plasmid was constructed as follows: the PstI fragment from pATHTNF, containing the complete coding sequence of the human tumor necrosis factor cDNA (hTNF), was blunted (Klenow exonuclease) and provided with BglII linkers. After treatment with BglII, the fragment was inserted into the unique BamHI site of pSV529. The orientation of the inserted fragment is sense relative to the SV40 late promoter.
The plasmid was designed for expression in SV40-permissive cells (CV-1, AP8) and especially in COS.
Since 39% of the SV40 genome is duplicated (early region), one should be aware of the possibility of deletions due to homologous recombination.
The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727561.1.
