Further information: The BamHI-ClaI fragments from pATgMIFNG were ligated in the unique BamHI and unique ClaI sites of pAT153. This resulted in two intermediate plasmids: pATgMIFNG28 containing exon 2, 3, 4 and the 3' untranslated region of the mIFNG gene (mIFNG) and pATgMIFNG2 containing the mIFNG 1st exon and the 5' untranslated region. In the unique PvuII site of pATgMIFNG2 a HindIII linker was added, resulting in pATgMIFNG2H3. A triple ligation was then realised: the HindIII-ClaI fragment of pATgMIFNG2H3 and the ClaI-BamHI fragment of pATgMIFNG28 were inserted in the HindIII-BamHI opened pSV23 vector.
Since the nucleotide sequences of the introns of the mIFNG gene could not be completely traced back, there is uncertainty as to the cutting frequency of the restriction enzymes, except for the sites that were analysed in the authenticity test.
The nucleotide sequence of the mouse IFNG cDNA corresponds with the EMBL Nucleotide Sequence Database accession number K00083.1.

Authenticity test: Restriction enzyme pattern analysed at GeneCorner: BamHI/HindIII, EcoRI, HindIII and PstI. The approximative length of the plasmid and of the mIFNG insert deduced from the compiled nucleotide sequence has thus been confirmed. Furthermore, the results revealed that PstI and EcoRI each cut at least 6 times. The locations of the extra PstI and EcoRI sites have not been determined; consequently, the compiled nucleotide sequence has not been adapted.
